摘要 目的探讨小分子化合物J2在抑制小鼠皮肤移植排斥反应的作用。方法以C57BL/6小鼠为供体,以BALB/c小鼠为受体建立皮肤移植实验模型,随机分为A、B、C组,并分别以CsA、J2、空白液连续ig12d为干预手段,比较各组移植皮瓣的存活时间和存活率,术后14d对各组做组织学检查。结果J2组发生排斥时间延迟,同种异体移植对照组皮瓣平均存活时间为(13.7±1.4)d,J2组为(17.7±2.4)d,两组差异有统计学意义,CsA组存活时间为(18.2±2.1)d,与J2组比较存活时间、存活率差异均无统计学意义。组织学检查证实,术后14dJ2组植片未见明显的淋巴细胞浸润。结论J2有抑制小鼠皮肤移植排斥作用。 Aim To investigate the effects of J2 on prevention of skin allograft rejection in mouse skin transplantation.Methods The C57BL/6 and BALB/c were used as donors and recipients to establish a skin allograft model, and dividedinto 3 groups randomly. Group A, allograft control (this group was given placebo only) ; group B and group C, allograftgroups,were treated with orally CsA and J2,respectively.The drugs were delivered for 12 days from the day of transplantation. Graft survival was assessed by Kaplan-Meier analysis. Fourteen days after transplantation, histological examinationwas performed to confirm the clinical diagnosis of rejection. Results The average transplant survival time in the allograftcontrol (Group A) was (13.7±1.4)d. Treatment with J2 (Group C) led to a statistically significant prolongation oftransplant survival to (17.7±2.4)d ( P〈0.01),but compared with treatment with CsA (Group B) (18.2±2.1)d,there was no statistically significant difference between the two groups( P 〉 0.05). Fewer inflammatory cells were foundin the skin allograft of J2 group. Conclusion These results showed that J2 could prevent skin graft rejection.
