摘要
目的:观察按摩内关穴加川芎嗪治疗对急性心肌缺血大鼠心肌、血清超氧化物歧化酶活力、丙二醛含量及血清乳酸脱氢酶、肌酸激酶活性的影响,并与单纯川芎嗪治疗结果进行比较,分析中医按摩对中药有效成分的靶向性影响。方法:实验于2005-09/11在河北医科大学中医学院中药药理实验室完成。①选用SD大鼠40只,雄性,鼠龄3月,体质量180～220g。按随机数字表法将40只大鼠分为4组,每组10只:正常对照组、模型组、川芎嗪组及川芎嗪+按摩组。②川芎嗪组和川芎嗪+按摩组:按20mg/kg剂量连续腹腔注射盐酸川芎嗪注射液[山西康意制药有限公司,批号040509,2mL(40mg)/支],5mL/kg,1次/d,共3d。正常对照组及模型组腹腔注射给予等量蒸馏水,1次/d,共3d。末次给药后30min,各组大鼠舌下静脉注射垂体后叶素(20U/kg)致大鼠急性心肌缺血,同时川芎嗪+按摩组:将大鼠固定,用自制的圆针(头圆钝,直径0.6mm,长5cm,柄长1cm,不锈钢)点按内关穴10min,用中等力度,以大鼠肢体微微颤动,但不鸣叫为度。③于注射垂体后叶素后60min。摘取眼球取血,分离血清,按照南京建成生物工程研究所提供的相应试剂盒说明书方法测定血清超氧化物歧化酶活力、丙二醛水平,血清乳酸脱氢酶活性、肌酸激酶活性。④处死大鼠,取出心脏,制成匀浆,按照南京建成生物工程研究所提供的相应试剂盒说明书方法测定心肌超氧化物歧化酶活力、丙二醛含量。⑤组间计量资料差异比较采用方差分析。结果:大鼠40只均进入结果分析。①模型组大鼠血清超氧化物歧化酶活力明显低于其他3组(P<0.05～0.01),血清丙二醛水平明显高于正常对照组和川芎嗪+按摩组(P<0.01),川芎嗪组血清丙二醛水平与模型组相近。②模型组大鼠心肌超氧化物歧化酶活力明显低于正常对照组和川芎嗪+按摩组(P<0.01,0.05),川芎嗪组大鼠心肌超氧化物歧化酶活力与�
AIM： To observe the effect of massaging Neiguan acupoint combined with ligustrazine on myocardium, activity of serum superoxide dismutase （SOD）, content of malondialdehyde （MDA）, activity of serum lactate dehydrogenase （LDH） and activity of creatine kinase （CK） of rats with acute myocardial ischemia, and compare the effect of single ligustrazine to analyze target-synergistic action of massage on effective components of Chinese herbs.
METHODS： The experiment was completed at Pharmacological Laborato- ry of College of Traditional Chinese Medicine of Hebei Medical University from September to November 2005.① Totally 40 male SD rats, aged 3 months, weighting 180-220 g, were selected in this study. All 40 rats were divided into 4 groups according to randomly digital table： normal control group, model group, ligustrazine group and ligustrazine ＋ massage group with 10 in each group. ② Ligustrazine group and ligustrazine ＋ massage group： 5 mL/kg hydrochloric ligustrazine injection with dosage of 20 mg/kg were injected intravenously once a day for 3 days. The injection was prov ided by Shangxi Kangyi Pharmaceutical Company Limited [batch number： 040509, 2 mL （40 mg）/tube]. Rats in normal control group and model group were injeeted with the same volume of distilled water once a day for 3 days. Thirty minutes after the last administration, 20 U/kg pituitrin was injected into sublingual vein so as to induce acute myocardial ischemia, meanwhile, rats in ligustrazine ＋ massage group were fixed and acupunctured at Neiguan with self-make retmd--dike needle （tip of needle materialize as stainless steel was round and dull with 0.6 mm in diameter, 5 cm in length and 1 cm in length of lug） for 10 minutes. Moderate stress was used to shiver limbs lightly but rats did not called. ③ Sixty minutes after injection of pituitrin, eyeball was selected to collect blood and separate serum.
