山羊MSTN基因EcoRV酶切多态性分析 被引量:1

摘要 采用限制性内切核酸酶EcoRV对山羊MSTN基因的676 bp扩增产物进行了PCR-RFLP多态性分析,检测到3种基因型,其酶切位点分别由2种共显性基因控制;对该片段的纯合型(AA、BB)分别测序发现,BB型在3783位的T→C。首次试验证实山羊MSTN内含子2(676 bp)区域存在多态性。 MSTN gene was amplified and an uniform fragment of 676 bp was obtained in goat. The 676bp PCR product was digested with restriction endomuclease EcoRV and genetic polymorphism was investigated by PCR - RFLP. Three kinds of genotypes were found in these populations,which were controlled by two alleles. The amplified fragments of AA and BB of the fragment were sequenced. The result showed that in 676 bp BB genotype there was one substitution mutation at the 3 783 site(T→C). The result above - mentioned first confirmed that there were polymorphisms in intron 2 (676 bp) region of MSTN gene.
