摘要
目的:探讨热激蛋白(HSP)的高表达和细胞凋亡在兔动脉粥样硬化斑块中的作用。方法:取兔动脉粥样硬化易损斑块28例,镜下将斑块分为4个区,分别为纤维帽区、脂核区、肩部区和中膜平滑肌区;采用免疫组化技术检测各区不同细胞HSP70的高表达情况、原位末端标记法(TUNEL)检测各区不同细胞凋亡情况。结果:纤维帽区与肩部区HSP70的表达阳性炎性细胞数均显著高于脂核区、中膜平滑区,纤维帽区平滑肌细胞HSP70的阳性表达显著高于其他3个区。脂核区TUNEL阳性细胞数明显高于其他3个区。纤维帽区与肩部中表达HSP70的阳性细胞数大于TUNEL阳性细胞数(P<0.05,P<0.01);脂核区内TUNEL阳性细胞数明显大于表达HSP70的阳性细胞数(P<0.01)。结论:易损斑块内HSP70的高表达主要在纤维帽区及肩部区,而细胞凋亡主要在斑块脂核周围。
Objective:To investigate the roles of overexpression of heat shock protein (HSP)70 and apoptosis in rabbit's vulnerable plaques, Method: Under microscope, the vulnerable plaque is divided into four regions: fibrous cap region, shoulder region, lipid-rich core region and media-muscular region. Overexpressions of HSP70 were detected in different regions in 28 samples of vulnerable plaques from 30 rabbits with SP immunohistochemical technique, Apoptosis was detected with TUNEL, Result: The overexpression levels of HSP70 in inflammatory cells on fibrous cap and shoulder regions were significantly higher than that of other two regions. The overexpression level of HSP70 in smooth muscle cells on fibrous cap region was significantly higher than that of the other three regions. The amount of TUNEL positive cells on lipid-rich core region was much more than that of other three regions. The overexpression cells of HSP70 in fibrous cap and shoulder regions were significantly higher than TUNEL positive cells ( P 〈0.05, P 〈0.01 ). TUNEL positive cells were significantly higher than the overexpression cells of HSP〉 in lipid rich core region( P 〈0.01 ). Conclusion:The overexpression of HSP〉 mainly occurs in fibrous cap and shoulder regions of rabbit vulnerable plaque tissue. Apoptosis mainly occurs in the region of lipidrich core.
