摘要
目的更好地开展麻疹病原学监测,分离麻疹病毒(MV)。方法对1994年和2003~2005年在我省分离的MV结果进行分析。结果1994年我省采集了5份咽拭子送国家麻疹实验室分离到2株MV,分别为China94-2和China94-3;2003年仅从采集的36份咽拭子中分离到1株MV;2004年和2005年采用vero-slam细胞从采集的142份咽拭子中分离到23株MV,出疹后3d内采集的合格标本分离率为20.91%(23/110),而在尿液标本和眼分泌物标本中未分离到病毒。来自不同的市采集的咽拭子标本分离比例差别较大。所有分离到的MV(26株)全部经中和试验和逆转录-聚合酶链反应鉴定,再经核苷酸序列分析鉴定均为H1基因型。结论我省已经掌握了MV的分离和鉴定技术,分离比例较高;我省多年来流行的麻疹毒株均为H1基因型,与国内流行的优势基因型一致。
Objective To develop pathogenic surveillance on measles and to effectively isolate measles virus. Methods We analyzed the data of measles isolation in 1994 and from 2003 to 2005 in our province. Results Two wild-type measles strains named China94-2 and China94-3 were obtained from 5 swab samples of Hebei Province in China measles lab. We got another wild-type measles strain from 36 swab samples in 2003 in Hebei Province lab. Twenty-three wild-type measles strains were obtained from 142 swab samples in 2004 and 2005 in Hebei Province lab. The measles isolation rate was 20.91%(23/110) from 110 swab samples which was collected in 3 days after rash occurrence. No measles virus strain was colleeted from samples of urine and eye secretion. There was significant difference in the ratio of measles isolation from the swab samples among the different regions. Twenty-six wild-type measles strains had been confirmed by neutralization test and RT-PCR. These 26 strains belonged to HI genotype of eighth genetic group of wild-type measles virus. Conclusions We have grasped the technical ability of measles virus isolation and confirmation and got higher isolation ratio. The wild-type measles virus isolated from Hebei Province is of H1 genotype from 1994 to 2005, which is the same as the dominant genotype circulation in China.
