摘要 目的研究肿瘤坏死因子-α(TNF-α)对人恶性胶质瘤U251细胞核因子-κB(NF-κB)的诱导活化作用,并观察NF-κB抑制剂对TNF-α诱导U251细胞凋亡的影响。方法采用电泳迁移率变动分析法检测TNF-α对U251细胞NF-κB的诱导活化,并通过甲基噻唑蓝(MTT)法和流式细胞技术检测NF-κB抑制剂对TNF-α诱导U251细胞凋亡的影响。结果TNF-α对U251细胞NF-κB具有诱导活化作用,其作用效果具有时间依赖性。NF-κB抑制剂对TNF-α诱导的U251细胞凋亡具有明显的促进作用(P<0.01)。结论TNF-α对胶质瘤细胞中的NF-κB具有诱导活化作用,抑制NF-κB活化可以提高胶质瘤细胞对TNF-α诱导的凋亡作用的敏感性。 Objective To study tumor necrosis factor-α (TNF-α) induced activation of nuclear factor-κB (NF-κB) in U251 human malignant glioma cells and to observe the influence of inhibitor of NF-κB on TNF-α induced apoptosis of U251 cells.Methods The activation of NF-κB induced by TNF-α in U251 cells was examined by electrophoretic mobility shift assay. The influence of NF-κB inhibitor on TNF-α induced apoptosis of U251 cells was examined by methyl thiazolyl tetrazolium (MTF) and flow cytometry (FCM) method. Results TNF-α could induce activation of NF-κB in U251 cells with a time-related fashion. TNF-α induced apoptosis of U251 cells were obviously enhanced by NF-κB inhibitor (P〈0.01).Conclusion TNF-α leads to the activation of NF-κB in glioma cells and the inhibition of NF-κB activation can increase the sensitivity of glioma cells to TNF-α induced apoptosis.
