摘要
目的研究正己烷对周围神经损伤的特征。方法60只SD大鼠灌胃给予正己烷0、168、675、2700mg/kg,连续8周(5d/周)。分别在0、2、4、8周末进行尾部混合神经传导速度的测定。实验结束后,处死部分动物,制作胫神经后部肌支束光镜、电镜切片,观察其变化。结果各剂量组混合神经传导速度(MNCV)减慢表现明显[P<0.05(低剂量4周),P<0.01(各剂量其余周次)],随染毒时间延长、剂量增加,MNCV损伤效应加重。胫神经光镜下表现为髓鞘数目稀疏、部分髓鞘大小不一,电镜下髓鞘形态各异,轴索肿胀、脱落、细胞器增多,神经丝密度增加。结论正己烷明显可致周围神经的功能和病理改变,其致周围神经病的病变类型为远端型轴索病(NF聚集型)。
Objective To analyse the specialities of peripheral neuropathy in rats induced by n-hexane. Methods Sixty SD rats were given 0,168,675,2 700mg/kg n-hexane for 8 weeks (5d/w) by garage. The mixed nerve conduction velocity (MNCV) was measured before and at the 2^nd, 4^th, 8^th week of administration with the tail nerve. The muscle branch of the posterior tibial nerve was observed under light microscope and transmission electron microscope after 8 weeks of exposure. Results Decreased MNCV were significantly observed [ P 〈 0.05 ( 168 mg/kg dosage, 2^nd week), P 〈 0.01 ( other dosages and other weeks) ] , MNCV impairment became much severe with longer-time and higher-doses, in the tibial nerve, morphological changes such as axons decreased in numbers and dissimilar in size were observed under light microscope. Diversiform in myelin sheath, swollen, demyelination,organelle hyperplasia in the axon and high dense malaligned neruofilaments were shown under transmission electron microscope. Conclusion The changes were significant in peripheral nerve's function and pathology. The type of neuropathy was peripheral distal axonopathy( accumulation of neurofilaments).
