摘要 目的研究针对c-myc基因的小干扰RNA(small interfering RNA,siRNA)对白血病细胞株HL-60细胞增殖及c-myc蛋白表达的影响,探讨应用siRNA进行白血病基因治疗的可能性。方法制备特异性对应c-myc基因的siRNA转染HL-60细胞,倒置显微镜观察细胞形态学改变,MTT法检测细胞增殖状态,免疫细胞化学染色法检测c-myc蛋白表达的改变。结果siRNA转染HL-60细胞后,细胞生长受抑,增殖能力减弱,其增殖抑制作用于转染后48h、72h最明显,增殖抑制率分别为53.2%和51.5%;免疫细胞化学染色结果显示c-myc蛋白阳性率从空白组(76.67±4.35)%下降至(23.33±3.56)%。结论针对c-myc基因的siRNA对HL-60细胞增殖及c-myc蛋白表达有明显抑制作用,有望为白血病提供新的治疗途径。 Objective To study the effects of small interfering RNA (siRNA) targeting c-myc gene on the proliferation and on the expression of c- myc proteion in HL-60 cells of leukemic cell line and to explore the gene therapeutic possibility of siRNA for leukemia. Methods The siRNA for c- myc gene was designed and transfected into HL-60 cells line, The morphological change was observed under inverted microscope, The cell proliferation was investigated by MTT method. Immuno-histochemistry was adopted to examine the expressions of c-myc proteion. Results After the siRNA was transfected, the growth of cells was repressed and the ability of proliferation decreased, the siRNA acted as a specific growth inhibitor to HL-60 cell,at 48h,72h after transfection, this inhibitory effect was marked, the rate of inhibition was 53.2% and 51.5 % respectively. The levels of c-myc protein after treatment decreased from (76.67 ± 4.35 )% to (23.33 ± 3.56)%, Conclusions The siRNA targeting c-myc gene could inhibit the proliferation of HL-60 cells obviously by inhibiting the expression of c-myc protein and might be a new therapeutic modalities in the treatment of leukemia.
