摘要
目的探讨小剂量低分子肝素钠(12.5U/ml)能提高兔骨髓间充质干细胞(BM-MSCs)在血管内皮生长因子(VEGF)诱导下时细胞增殖能力和贴壁率。方法联合应用肝素和VEGF共同作用于兔骨髓MSCs,比较肝素组和未加肝素组细胞的生长曲线和贴壁率曲线,并观察细胞的形态变化。结果肝素组诱导细胞的倍增时间为(47.3±3.7)h,未加肝素组为(53.6±6.5)h,P=0.000<0.01;肝素组贴壁率较无肝素组稍微降低,P=0.921>0.05。两组细胞形态都呈“铺路石状”,差别不大”。结论小剂量低分子肝素钠能够增强兔骨髓MSCs在VEGF作用下增殖能力。
Objective To investigate whether LMWH(low molecular weight heparin)can promote proliferation of rabbit MSCs (Mesenchymal stem cells)under VEGF in vitro. Methods MSCs is cultured in the medium including VEGF and LMWH. The morphology of the cell is observed under optic microscope,and investigated the proliferation ratio by using MTr-cell proliferation detection and adherence ratio.Results The doubling generation time of heparin group: (47.3 ± 3.7) h, the non-heparin group: (53.6 ± 6.5 ) h, the pared-samples T teat shows P = 0.000 〈 0.01 ;the adherence ratio of heparin group decrease slightly,and the K. Pearson x^2 test shows P = 0.921 〉 0.05;both groups are cobbe-stone-like. Conclusion Low dose LMWH can remarkably promote the proliferation of rabbit MSCa under VEGF in vitro.
