摘要
昆明鼠卵母细胞hCG注射后18 ̄19h用体积分数为7%的乙醇作用5 ̄6min,能够获得较为理想的激活效果,激活率高达90%。乙醇激活处理时,卵丘细胞以及细胞外Ca\+\{2+\}、Mg\+\{2+\}的有无对卵子的激活效果均无显著影响。37℃下激活处理与30℃下相比,激活率无显著差异,但卵子碎裂率显著提高。性成熟前小鼠超排的卵子虽然也能被激活,但激活率低于成熟后小鼠超排的卵子。哺乳动物卵母细胞成熟分裂抑制剂NaF和IBMX不能抑制乙醇处理卵的孤雌激活,但显著地抑制了激活卵子的卵裂。
Kunming mouse oocytes(18 19 h post hCG) could be activated efficiently by treatment with 7% (V/V) ethanol for 5 6 min, and the rate of activated oocytes was about 90%. The parthenogenetic activation rate was not affected by cumulus cells and Ca 2+ , Mg 2+ in activated medium. The ethanol activation rate at 37℃ was as high as that at 30℃, but the fragment rate at 37℃ was higher than that at 30℃. The oocytes removed from mice before sexual maturity could also be activated, but the activation rate was lower than that of oocytes removed from sexual mature mice. NaF and IBMX failed to inhibit the parthenogenetic activation of ethanol treated oocytes, but reduced the cleavage rate of the activated oocytes significantly.
