阿仑膦酸钠对体外培养的间充质干细胞成骨能力的影响 被引量:2

摘要 目的探讨阿仑膦酸钠对于体外培养的人间充质干细胞分化与增殖的影响,并对其分子机制进行初步研究。方法体外分离与扩增人骨髓间充质干细胞,用不同剂量的阿仑膦酸钠处理72 h后,MTT法检测细胞的增殖能力,全自动生化分析仪分析碱性磷酸酶活性以反映成骨分化能力,RT-PCR检测各种重要成骨相关转录因子的表达情况。结果10-7M,10-8M与10-9M阿仑膦酸钠组的增殖速度明显大于对照组(P<0.05),而且碱性磷酸酶与骨钙素的表达量也明显增高(P<0.05),转录因子Runx2,Dlx5,Osx表达有所增高。结论阿仑膦酸钠能够同时促进人骨髓间充质干细胞的增殖与分化,其作用可能与成骨转录因子有关。
