C反应蛋白与肾小管间质纤维化关系研究

摘要 [目的]探讨C反应蛋白(CRP)与慢性环孢素A(CsA)肾毒性肾小管间质纤维化的关系.[方法]给雄性Sprague-Dawley大鼠皮下注射CsA 4周,建立慢性CsA肾毒性动物模型.采用胶乳增强免疫比浊法、免疫比浊法以及免疫组织化学染色方法检测对照组及慢性CsA肾毒性组大鼠血清高敏CRP(hs-CRP)、CRP及肾内CRP的水平;采用三色染色方法观察肾小管间质纤维化程度;以直线相关分析法对CRP与肾小管间质纤维化程度间的关系进行分析.[结果]慢性CsA肾毒性组与对照组血清CRP和hs-CRP的含量水平间无显著性差异,与肾小管间质纤维化程度无相关关系;慢性CsA毒性组大鼠肾内CRP的表达显著高于对照组,与肾小管间质纤维化程度呈正相关(r=0.862).[结论]慢性CsA肾毒性肾小管间质纤维化与肾内CRP的表达水平密切相关,与血清CRP的含量无关. OBJECTIVE To study the relationship between C-reactive protein (CRP) and tubulointerstitial fibrosis (TIF) in a model of chronic cyclosporine (CsA) nephrotoxicity. METHODS Chronic CsA nephrotoxicity was induced in Sprague-Dawley rats by administering CsA for 4 weeks. Serum CRP, high sensitivity-CRP (hs-CRP), and intrarenal CRP of rats in control group and CsA treatment group were evaluated with particle-enhanced immunoturbidometric method and immunohistochemistry. TIF was measured by Masson trichrome stain. The relationship between CRP and TIF was analyzed by the Pearson single-correlation coefficient. RESULTS There were no significant differences in CRP and hs-CRP levels between two groups Moreover, there was no relationship between serum CRP and TIF, intrarenal CRP expression was markedly increased in CsA-treated rat kidneys compared with control, and that was correlated with the TIF score ( r = 0. 862), CONCLUSION Intrarenal CRP level but not serum CRP is closely associated with TIF in chronic CsA nephrotoxicity.
