摘要
根据小鼠α-1,3-GT基因mRNA序列设计并合成了特异性的发夹状RNA干扰序列,以pCDNA3为基本骨架,选取人类H1启动子,构建了针对小鼠-α1,3-GT基因的特异性基因沉默载体,为进一步获得α-1,3-GT基因沉默小鼠,构建异种器官移植用小鼠模型奠定了基础。
RNAi method has now become a powerful tool to study gene function. In this paper, we describe the construction of siRNA expression vector targeting the α-1,3-GT gene of mouse. Firstly, two siRNA sequences are designed basing on the mRNA of mouse α-1,3-GT gene. Then, the designed siRNA sequences and the necessary sequence of human HI promoter are synthesized. The final vector is completed by ligating siRNA sequence and H1 promoter into backbone of pCDNA3. This will constitute a important foundation for further development of α-1,3-GT knock-down mice and for establishment of mouse model for xenotransplantation research.
