导数-同步荧光法测定三月泡叶中总黄酮含量的研究 被引量:8

摘要 黄酮类化合物能与Al^3+形成稳定的荧光配合物,依此建立了同步荧光法测定三月泡叶中总黄酮的含量的新方法。用80%乙醇加热回流提取三月泡叶中总黄酮,以芦丁为标样,选择盐为40nm,采用导数-同步荧光光度扫描测定三月泡叶中总黄酮含量,并进行加标回收实验,验证方法的准确度和可靠性。结果表明,本方法检出限为1.27×10^-9mol·L^-1,建立的线性回归方程为y=0.2925x+1.1605,其相关系数r=0.9984,线性范围在0-3.3087×10^-5mol·L^-1之间,平均回收率0.9984。相对标准差(RSD)为0.84%。本方法操作简便、快速、准确,适应于各类可与金属离子形成荧光配合物之天然组份的快速分析。 On the basis of reaction of total flavanoid with alumimum ion to form synchronous fluorescent materal, the new method of determining total flavanoid in leaves of Rubus corchorifolius L.f was established.Using 80% ethanol by continuous themml reflux to extract the Rubus corchorifolius L. f leaves, taking rutin as standard sample, selecting △λ = 40 nm and scanning by derivative- sychronous fluorometry, detenning content of total flavanoid, then inspecting the accuracy by recovery test.The detectiong limit was 1.27 ×10^-9mol·L^-1, the range of linearity with concentration of rutin was between 0 - 3.3087 ×10^-5mol·L^-1, linearity relationship y = 0.2925x + 1. 1605, correlativity 0.9984, RSD 0.84% .The method is fit for analysis of natural material which can be formed fluorescent complex compound with metal ion.
