摘要
了解肺炎克雷伯菌对庆大霉素等氨基糖甙类的耐药机制。收集1990-1994年分离的庆大霉素耐药菌132株,用药敏分析、分析杂交及接合转移试验,调查耐药基因的分布及可能的扩散机制。
Objective To understand the resistance mechanism of K.pneumonia to gentamicin. Methods Wecollected 132 isolates resistant to gentamiein,investigated the distribution and probable mechanisms of spreading of gentamicin resistance gene mainly by using techniques of molecular genetics. Results Most strains(116/132)contain homologous sequence of N-acetyltransferase(3)-Ⅱa gone. Moreover,it is located on an approximately 85×10~6 conjugative plasmid as well as chromosome.Con- clusions A transposon might carry gentamicin resistance gone and transconjugate experiment indicated that this large plasmid also carried penicillin,chloramphenicol and kanamycin resistance genes.All these antibiotics resistance genes were transferable to E'coli DH5α and HB802 by cojugation.MICs of 13 aminoglycosides on these isolates were determined by agar dilution methods.Most strains were resistant to gentamicin,tobramycin,netilmicin,2-netilmicin,6-netilmicin,ere,but susceptible to isepamicin and fortymicin.All were susceptible to aparamycin.
