摘要 目的:探讨生物可降解聚乳酸-聚乙醇酸-聚乙二醇(PLGA-PEG)血管支架置入对兔动脉粥样硬化模型的血清基质金属蛋白酶2,9(MMP-2、MMP-9)的影响。方法:成年健康白兔30只,经球囊血管内膜剥脱法制成髂动脉动脉粥样硬化动物模型,喂养6周后,随机分为两组,实验组(PLGA-PEG组,n=15),给予血管支架(PLGA-PEG)置入,对照组(n=15)常规喂养。两组动物在支架置入前、置入后7及30 d分别收集耳缘静脉血1 mL,以SDS-PAGE Zymography法检测MMP-2、MMP-9)及相应酶原(pro MMP-2和pro MMP-9)。结果:实验组支架置入前所测的MMP-2、MMP-9和proMMP-2[(11 568±2 219)、(10 364±1 429和(13 649±1 894)INT.mm2)]明显高于对照组[(6 128±1 562)、(8 519±2 167)和(8 413±2 156)INT.mm2](P<0.05);但proMMP-2两组比较,差异无显著性(P>0.05)。实验组支架置入后7 d与置入前相比,MMP-2及MMP-9以及相应酶原虽有增高趋势,但差异无显著性(P>0.05);支架置入30 d后MMP-2及酶原高于置入前[(13 935±2 167)、(15 628±1 739)INT.mm2vs(11 568±2 219)、(13 649±1 894)INT.mm2](P<0.01),MMP-9及酶原亦增高,但差异无显著性(P>0.05)。结论:血清MMP-2及酶原在动脉硬化动物模型中明显增高,血管支架置入后MMP-2增高。 Objective To investigate the effects of polylactic acid-polyglycolic acid-polyethylene (PLGA-PEG) stent on matrix metalloproteinase (MMPs) in rabbits with atherosclerosis. Methods 30 adult healthy rabbits were divided randomly into two groups: control group (n= 15) and PLGA-PEG group (n= 15). Right iliac arteries of rabbits of PLGA-PEG group were injured by balloon denuded endothelization. PLGA-PEG stents were implanted in rabbits in PLGA-PEG group. MMP-2, 9 and pro MMP-2, 9 levels of PLGA-PEG group were determined before implantation, 7 and 30 d later by SDS-PAGE zymography. MMP-2, 9 and pro MMP-2, 9 levels were determined in control groups. Results The concentrations of MMP-2, 9 and pro MMP-2 in PLGA-PEG group were much higher than those in control group (11 568±2 219, 10 364+1 429, 13 649+1 894 INT ·mm^2 vs 6 1285=1 562, 8 519+2 167, 8 413+2 156 INT ·mm^2 , P〈0.05). There were no significant differences of pro MMP-9 between two groups (P〈0.05). In PLGA-PEG group, the levels of MMP-2, 9 and pro MMP-2, 9 didn't elevate significantly 7 d later (P〈0.05). However the MMP-2 and pro MMP-2 concentrations were still higher 30 d after implantation (13 935±2 167, 15 628±1 739 INT·mm^2 vs 11 568±2 219, 13 649±1 894 INT ·mm^2, P〈 0.01). Conclusion The serum levels of MMP-2 and pro MMP-2 increase significantly in rabbits with atherosclerosis. The MMP-2 level also increases after implantation.
