摘要
叙述了肉中伊维菌素残留量的柱前衍生HPLC分析方法。牛、猪肉中的伊维菌素用乙腈提取后,经C18固相萃取柱净化,蒸干净化液后,再用三氟乙酸酐和N-甲基咪唑衍生,在高效液相色谱仪C18柱上以100%甲醇为流动相,在激发波长364nm,发射波长470nm条件下,进行分离并经荧光检测。结果表明:伊维菌素浓度与峰面积在0.01~2.00μg/mL(0.05~10.00ng)范围内有很好的线性关系;检出限为2.656×10-5mg/kg;0.04mg/kg水平的添加试验的平均回收率为100.5%,变异系数是1.3%;用该法对乌鲁木齐市场牛肉4个样品和猪肉5个样品进行了实际测定。
A HPLC method was developed and studied. The Ivermectin in beef and pork was extracted by acetonitrile, purified by C18 Column of solid-phase extraction (SPE), then derived by trifluoroacetic anhydride and 1-methylimidazole.And the samples were measured reverse phase HPLC with fluorescence detector. The method showed good results: the linearcorrelation coefficient was 0.99999 between the range of 0.01 -2.00μg/mL(0.05 - 10.00ng), the detection limit was 2.656×10^-5 mg/kg, the average recoveries of Ivermectin in the beef and pork added in 0.04mg/kg were 100.5%, the coefficient of variability was 1.3%. New method was used to determinate several related market samples.
