摘要
采用缺陷型逆转录病毒转导人TNF-α基因至口腔癌DNL,并应用PCB技术、TNF-α活性检测技术、G418筛选试验对DNL中外源基因及其上清液中的TNF-α活性进行了检测。结果:1.转导基因的DNL中有外源基因存在;2.转导基因的DNL上清液中有高活性的TNF-α;3.转导基因DNL能在G418培养基中生长。表明缺陷型逆转录病毒载体能够将TNF-α基因导入口腔鳞癌DNL中表达,转导基因DNL能够分泌高活性的TNF-α。该结果为在口腔癌领域开展细胞因子基因治疗奠定了基础。
The gene coding for TNF-α was introduced into DNL of oral squamous cell carcinoma with retroviral-mediated gene transduction,TNF-α gene and TNF-α production was detected.The results showed:1.With PCR technique,exogenous gene found in transduced DNL;2.TNF-α production was found in supernatant of transduced DNL;3.Transduced DNL could grow in G418 medium. The test demonstrated that the gene coding for TNF can be introduced into DNL of oral squamous cell carcinoma with retfoviral-mediated gene transduction.This study suggest that TNF-α gene transduction may be important strategic approach to therapy of oral cancer.
