参照GenBank中布氏杆菌的全基因组序列,设计一对特异性引物,通过PCR方法,扩增了8株来自3个布氏杆菌种的布氏杆菌表面蛋白31(BCSP31)全基因。PCR产物经克隆和序列分析后发现,该基因非常保守,8株不同菌株间的同源性高于99.3%;进化关系分析显示,5株中国来源的菌株(S2,M5,M111,M28,A387)显示了最近的同源关系,其中2个弱毒菌株S2和M5的BCSP31基因序列100%同源。3株外国来源的菌株(S19,2308,Rev.1)中,S19和2308同源性为100%。Rev.1与其他7株布氏杆菌BCSP31基因均有较大的差异。序列分析结果表明,BCSP31基因序列差异与布氏杆菌的种属和毒力无相关性。
A pair of primers was synthesized for amplification of brucella cell surface protein 31 ku ( BCSP31 ) gene according to the complete genomic sequence of Brucella abortus published in GenBank. BCSP31 genes were amplified from 8 Brucella strains belonging into 3 different species by polymerase chain reaction (PCR). The PCR products were cloned in pMD18-T vector for sequencing. Sequence analysis showed that BCSP31 genes were conservative, and the identical percent of these genes was more than 99.3% between these 8 strains. Evolution analysis revealed that 5 Chinese strains ( S2, MS, M111, M28, A387 ) had the nearest relationship, among them 2 attenuated strains, S2 and MS, had the same sequences in BCSP31. Among the 3 foreign strains(S19,2308, Rev. 1 ), there was only little difference between S19 and 2308 in BCSP31 sequence. Rev. 1 was significantly different not only from the 5 Chinese strains, but also from S19 and 2308. No relationships were found between the sequence divergence of BCSP31 gene and the species and virulence of Brucellas.
