摘要
应用鲎试剂动态浊度法,将硫酸链霉素制备成4、2、1、0.5mg/mL溶液,定量添加标准内毒素进行干扰预试验,计算回收率,确定不干扰浓度为0.5—2mg/mL。从中选出1mg/mL为最佳浓度,然后进行3个批次硫酸链霉素正式干扰试验。在供试品中定量添加标准内毒素,其回收率分别为125.8%、126.1%、127.3%,均在50%-200%之间,说明此浓度的供试品对鲎试剂反应无干扰作用。因此,内毒素含量在0.015—1EU/mL范围内,将硫酸链霉素制备成浓度为1.0mg/mL的溶液,可用于细菌内毒素的定量检测。
In pre-interference experiments, the amount of endotoxin was added to 4 different concentrations of streptomycin sulphate at 4,2,1 and 0.5 mg/mL respectively. The recovery rates were calculated. It showed that the concentration 0.5 - 2.0 mg/mL of streptomycin sulphate did not interfere with limulus test. It was concluded that 1 mg/mL of antibiotic was the optimum concentration. On this condition, the interference experiments were conducted in three batches of streptomycin sulphate (at 1 mg/mL) and quantitative volumes of standards endotoxin were added, the recovery rate was 125.8%, 126.1% and 127.3%, respectively. The recovery rates of endotcoxin in 3 batchs streptomycin sulphate were in the range of 50% - 200%. As a conclusion, the content of endotoxin were in the range of 1,0.25, 0.625 and 0. 015 EU/mL, and the streptomycin sulphate solution of 1 mg/mL did not interfere with limulus test. The dynamic turbidimetric limulus test could be conveniently used for the endotoxin assay in streptomycin sulphate.
