摘要
为探讨胃内局部环境中胃酸、胃蛋白酶等因素对胃安素(TP)药效的影响,对经人工胃液处理前后三批不同时间制备的TP进行了活力检测及动物模型试验。结果:1.未经人工胃液处理,三批TP(100mg/L)均显示较强的促3T3成纤维细胞增殖活力,而以920620及930713批TP偏低。2.经人工胃液处理0.5、1、2小时,930713、940528二批TP均有活力,以940528批活力较高,即经处理1小时、稀释度为2mg/L时,活力比值X>2.0;而920620批未能测出活力(X<2.0)。3.动物模型实验结果:与对照组比较,930713、940528批TP组溃疡面积在喂药10天后均缩小,具高度显著性差异(P<0.01);920620批TP疗效则不显著(P>0.05)。结果提示,人工胃液条件下pH、胃蛋白酶对TP活力有一定影响,并与其消化作用及TP储存时间长短相关。活力检测与动物模型试验结果相符,并具一定的平行性。
Thymic Protein(TP) is a soluble extract prepared from animal thymus. Three different lots of TP were treated by artificial gastric juice (AGJ)in order to investigate the impact of factors such as gastric acid and pepsin on the pharmacodynamic action of TP-The results showed as following:1. Before the AGJ treatment .all the 3 samples of TP(100mg/L) of 3 lots had significantly high activity of promoting the 3T3 fibroblast proliferation. 2. TP of 930713 and 940528(lot number)had the activity ratio X>2. 0 at the time intervals of 0. 5,1 and 2 hr. after AGJ treatment,while 920620 lost its activity. The activity of 940528 was the highest, maintaining its high activity 1 hr. after AGJ treatment at the TP concentration of 2 mg/L. 3. The ulcer area in all the treatment group with TP of 930713 and 940528 were diminished at the 10th day after the operation, the results possessing very significant difference(P<0. 01) compared with that of the NS control,with exception of that with TP of 920620. The results suggested that pH and pepsin may affect the pharmacodynamic action of TP under the AGJ condition,the effects were closely correlated with the time interval of TP storage and AGJ digestion.
