糖皮质激素对活化后小胶质细胞株N9中MHC-Ⅱ和OX40L表达的影响 被引量:6

摘要 目的研究糖皮质激素对炎症刺激活化后的小胶质细胞株N9表面分子MHC-Ⅱ和OX40L表达的影响。方法脂多糖(LPS)联合小鼠γ-干扰素刺激N9细胞活化,采用RT-PCR、免疫荧光检测N9活化前后OX40L的表达。糖皮质激素及其受体阻断剂RU-486处理活化的N9细胞后,用流式细胞术(FACS)检测MHC-Ⅱ、OX40L的表达变化。结果N9活化前OX40L mRNA和蛋白均有组成性表达,活化后MHC-Ⅱ和OX40L表达显著上调(P<0.05),糖皮质激素处理后与活化组比较两分子表达下调(P<0.05),加入RU-486又可使两分子表达上调。结论小胶质细胞N9活化后MHC-Ⅱ、OX40L表达显著升高,而糖皮质激素可通过抑制小胶质细胞MHC-Ⅱ和OX40L表达拮抗小胶质细胞的抗原呈递功能,从而降低由T细胞活化介导的损伤效应。 Objective To investigate glucocorticoid effects on the expressions of MHC-Ⅱ and OX40L in activated mieroglia cell line Ng, Methods N9 were activated by the stimulation of mouse-interferon-gamma (m- IFN-γ) and lipopolysaceharide (LPS), Expressions of OX40L were observed by RT-PCR and immunofluorescenee. Glueoeorticoid and RU-486 were added to the activated N9 , expressions of MHC-Ⅱ and OX40L were analysed by FACS. Results OX40L mRNA and protein were constitutively expressed, and the expressions of MHC-Ⅱ and OX40L were increased in the activated N9, while glueocorticoid down-regulated their expressions. These changes were reversed by the glucoeorticoid receptor blocker RU-486. Conclusions Expressions of MHC-Ⅱ and OX40L were increased obviously in the activated Ng, and glueocorticoid could inhibit mieroglial functions of antigen presentation by suppressing their expressions, and to reduce T cell-mediated damages,
