摘要
目的 研究腺苷三磷酸酶(p97)在非小细胞肺癌(non-small cell lung carcinoma,NSCLC)组织中的表达及与侵袭、转移的关系。方法 采用免疫组织化学和Western-Blot方法对60例NSCLC、癌旁组织以及10例正常肺组织进行D97蛋白定位表达及表达水平研究。结果 在免疫组化试验中,p97在NSCLC组织中的阳性表达率为65.0%(39/60),瘤旁组织为28.3%(17/60),两者比较差异有统计学意义,X^2=6.1,P=0.047;在Western印迹实验中,p97在NSCLC组织中的阳性表达率为81.7%(49/60),瘤旁组织为35.0%(21/60)。非肿瘤肺组织为10%(1/10),差异有统计学意义。X^2=7.5,P=0.023。Western印迹与免疫组化检查结果明显正相关性。r=0.293。P〈0.05。在NSCLC组织中,p97蛋白阳性表达率与肿瘤转移(P=0.0020)和肿瘤分期(P=0.0150)之间的差异有统计学意义;而与瘤体大小、病理分级、组织类型的差异无统计学意义。结论 NSCLC在蛋白水平上表达p97增加,与肺癌侵袭和转移有关。
Objective This study was designed to investigate the coorelation between the express of p97 with local infiltration and metastasis in Non = Small- Cell Lung Carcinoma (NSCLC). Methods Immunohistochemical and Westem Blot analysis for p97 were performed on samples of tumor tissue and the paratumor tissue from 60 NSCLC cases and 10 normal lung tissues. Results By immunohistochemical analysis, no specific positivity was detected in normal lung tissue,28.3% (17/60) in paratumor tissue and the tumor tissues showed a positive reaction to p97 about 65.0% (39/60) with significant difference, X^2 = 6.1, P = 0.047. The positive expression rate of p97 protein in tumor tissue determined by Western Blot analysis is 81.7% (49/60), 35.0% (21/60) in paracarcinoma tissue, and 10% (1/10) in normal lung tissue with significant difference, X^2 = 7.5, P = 0. 023. The positive rexpression of p97 protein determined by immunohistochemical analysis was similar to that by Western - Blot analysis. The expression rate of p97 protein in NSCLC with distant metastasis and high stage were significantly higher than that in NSCLC with low stage(P = 0.0150)or without distant metastasis(P = 0.0150). No correlation between p97 positive expression rate and other clinic pathological features in p97 negative or positive NSCLC cases was found.Conclusion In NSCLC,p97 overexpression at protein level may be closely related to the local infiltration and distant metastasis.
