HCV核心蛋白、突变p53和Bcl-2在HCC中的表达及相关性

摘要 目的探讨HCC组织中的核心蛋白、突变p53、Bcl_2的表达以及其相关性,探讨HCV核心蛋白是否促进p53突变和Bcl_2的表达。方法收集手术切除HCC组织42例,采用免疫组织化学EnVision法检测HCC组织核心蛋白、突变p53、Bcl_2的表达,用统计学分析三者间的关系。结果C蛋白、p53和Bcl_2在HCC癌组织中的表达率分别为40.5%(17/42)、47.62%(20/42)、83.3%(33/42);3组强度等级资料Kruskal_Wallis秩和检验H=16.33,差异性显著,Mann_WhitneyUtest:C蛋白和p53、Bcl_2蛋白间的P值分别0.43、0.00;C蛋白与突变p53、Bcl_2阳性强度两者间相关性检验P值分别为0.000、0.914,相关系数rs分别为0.67、0.08;突变p53与Bcl_2两者相关性检验P值为0.27,相关系数rp为0.32。结论3种蛋白的表达有相关性;HCV核心蛋白可能促进野生型p53突变和表达;核心蛋白和突变p53都有可能促进Bcb2蛋白的表达。 Objective To investigate expression of HCV core protein, mutant p53, Bcl-2 proteins and explore correlation of these three proteins expression in the tissues of HCC ; to explore wether HCV core protein promote wild p53 to mutation p53 and Bcl-2 protein expression. Methods Collect tissues of 42 cases with HCC and determine the expression of HCV core, p53, Bcl-2 proteins with immunohistochemical method, using Envision method;analyze the correlation of the three proteins by statistics. Results The positive expression of HCV core and mutation p53 proteins primarily expressed in the nucleus, while the positive expression of Bcl-2 protein in the cytoplasm; the positive rates of HCV core, mutation p53, Bcl-2 proteins were 40.5% ,47.62% ,83.3% respectively in the tissues of HCC;and so there was extremely difference (P = 0.000) in the three protein expression. While Mann-Whitney U test : p values of HCV core protein with mutant p53, Bcl-2 were 0.43,0.00 respectively;p values of correlationship test of positive intensity between HCV core and p53, HCV core and Bcl-2 were 0.000,0.914 respectively, while the correlation coefficient was 0.67, 0. 08 respectively; p value of correlation test of positive intensity between mutationp 53 and Bcl-2 was 0.27, and the correlation coefficient was rp = 0.32. Conclusion The expression of the three proteins is correlation : probably the HCV core protein promotes mutation of wild p53 and its expression, while HCV core maybe indirectly increase the expression of Bcl-2 protein, all of which may be contributed to hepatoeellular proliferation, transformation and immortalization.
