摘要
目的研究姜黄提取物(姜黄素)对大鼠肝星状细胞增殖和活化的影响。方法培养大鼠T6肝星状细胞株,并使用姜黄素对细胞进行处理,采用四甲基偶氮唑蓝[3-(4,5-dimthy-2-2thiazoly)2,5-dipheny-2H-tetrazoliun bromide,MTT]比色分析法检测药物对细胞增殖的影响;收集细胞并抽提细胞总蛋白,10%聚丙烯酰胺凝胶电泳分离蛋白,采用Western blot检测药物处理对细胞表达α平滑肌肌动蛋白的影响。结果姜黄素剂量依赖性地抑制大鼠T6肝星状细胞株的增殖,并可以降低细胞表达α平滑肌肌动蛋白的水平。结论姜黄素能够抑制肝星状细胞的增殖和活化.
Objective To study the Effect of Curcuma longa Linn. extraction (Curcumin) on the proliferation and activation of rat T6 hepatic stellate cell. Methods The rat T6 hepatic stellate ceils were cultured and treated by Curcumin at different concentrations. The inhibition effect was determined by MTT [3-(4, 5-dimthy-2-2thiazoly) 2, 5-dipheny-2H-tetrazoliun bromide] colorimetry. Cells were harvested and lysed to extract the total cellular proteins. Proteins were isolated by SDS-PAGE (sodium dodecyl sulfate- polyacrylamide gel electrohoresis, SDS-PAGE) and the expression level of α - smooth muscle actin after drug treatment was determined by Western blot. Results Curcumin inhibited the proliferation of rat T6 hepatic stellate cells in dose-dependent manner, reduced the expression of α smooth muscle actin. Conclusion Curcumin could significantly inhibit the proliferation and activation of rat T6 hepatic stellate cells.
