摘要
目的:建立新生大鼠缺氧缺血性脑病(HIE)模型,探讨降钙素基因相关肽(CGRP)对缺氧缺血性新生大鼠脑组织匀浆中内皮素(ET)的影响及CGRP对脑损伤的保护作用。方法:将7日龄新生大鼠分为3组,正常对照组(假手术组);生理盐水组给予生理盐水3μg/(kg·d),腹腔内注射,连续3d;药物治疗组给CGRP3μg/(kg·d),腹腔内注射,连续3d,断头取脑,用放射免疫分析法检测新生大鼠脑组织匀浆中ET的含量变化。结果:生理盐水组ET(112.55±20.70)pg/ml显著性升高,与正常对照组(53.02±12.39)pg/ml比较,差异有显著性P<0.01;药物治疗组ET(61.01±14.44)pg/mlCGRP与正常对照组比较,差异无显著性P>0.05。结论:ET参与缺氧缺血时的病理生理过程;CGRP能降低新生大鼠缺氧缺血脑损伤后ET的含量,CGRP对缺氧缺血脑损伤有保护作用。
Objective To establish Hypoxic-ischemic(HIE) model of infant rats and observe the changes of ETin brain tissue of infant rats with HIE. Methods Infant rats were distributed randomly into 3 groups with 15 infant rats for each group: normal control, salt group and HIE with given CGRP cured. Results The levels of ET of salt group were much higher than those of group normal control. The levels of ET of group given CGRP were declined, there was no significant with group normal control. Conclusion ET was involved in the development of hypoxic-ischemia in neonatal rats. CGRP could protect the neonatal rats from hypoxic-ischemie brain damage.
