水蛭注射液对大鼠脑缺血再灌注损伤神经细胞凋亡的影响 被引量:9

Effects of ShuiZhi Injection on Nerve Cell Apoptosis in Rats with Cerebral Ischemic Reperfusion Injury

摘要 目的:观察水蛭注射液对大鼠局灶性脑缺血再灌注损伤神经细胞凋亡的影响,探讨其神经保护作用机制。方法:选用健康Wistar大鼠60只随机分为假手术组,生理盐水对照组,川芎嗪对照组,水蛭注射液大、小剂量组,每组各12只。除假手术组外,其余四组均采用栓线法制作大鼠脑缺血再灌注损伤模型。分别于规定时间腹腔注射生理盐水(4ml/kg)、川芎嗪(6ml/kg)、水蛭注射液(2ml/kg或4ml/kg),进行神经功能评分,测定脑梗塞面积,计算脑组织神经细胞凋亡的百分率。结果:水蛭注射液大、小剂量组大鼠再灌注损伤后的神经功能评分、脑梗死面积百分比及脑神经细胞凋亡率与生理盐水对照组比较有显著性差异(P<0.01),尤以水蛭注射液大剂量组为明显。结论:水蛭注射液能改善大鼠脑缺血再灌注损伤的神经功能缺失,减少脑梗死面积,并能减低脑神经细胞凋亡的发生率,从而对脑缺血具有保护作用。 Objective: To observe the effects of ShuiZhi Injection on nerve cell apoptosis in rats with cerebral ischemic reperfusion injury, and explore its mechanism of nervous protective effect. Methods: 60 Wistar rats were randomly divided into the pseudo - operation group, the normal saline group, the ChuartXiongQing group, and the groups of high, low dose ShuiZhi Injection. Except the first group, the rest groups were made into the models by cork - line method, and were given abdominal injection with normal saline, ChuanXiongQing, and ShuiZhi Injection respectively to evaluate the nervous function, determine the cerebral infarction area, and calculate the rate of cerebral nerve cell apoptosis. Results: There were great differences of nervous function evaluation, cerebral infarction area, and rate of cerebral nerve cell apoptosis between the groups of high and low dose ShuiZhi Injection and the control group (P 〈 0.01), especially the group of high dose ShuiZhi Injection. Conclusions: ShuiZhi Injection can improve the nervous function in the rats with cerebral ischemie reperfusion injury, reduce the cerebral infarction area, and lower the rate of cerebral nerve cell apoptosis. It has the protective effects on cerebral ischemia.
