摘要
目的:研究复发性生殖器疱疹(RGH)患者外周血单一核细胞凋亡对Th1/Th2型细胞分化的影响。方法:分离RGH患者和健康体检正常人的外周血单一核细胞(PBMC),在PHA的刺激下体外培养48h后收集细胞和上清液,经流式细胞仪检测凋亡细胞,应用酶联免疫吸附试验检测培养上清液IL-2及IL-10。结果:RGH组新鲜血PBMC的凋亡率很低,与对照组相似(P>0.05)。培养48h后RGH患者的外周血单一核细胞凋亡明显高于正常对照组(P<0.001),RGH患者的培养上清液IL-2浓度明显低于对照组(P<0.001),而IL-10浓度显著高于对照组(P<0.001)。结论:PBMC凋亡异常严重影响了Th1/Th2型细胞分化,可能是导致复发性生殖器疱疹患者细胞免疫功能低下的重要机制之一,也可能在生殖器疱疹复发中起一定的作用。
Objective: To investigate apoptosis of peripheral blood mononuclear cells (PBMC) to effect of Th1/Th2 type cells proliferation in the recurrent genital herpes (RGH), Methods: Peripheral blood mononuclear cells (PBMC) were isolated from normal healthy physical examiination's persons (normal control group) and patients with RGH. PBMC were cultured with PHA - P for 48h in vitro. Apaptosis of the PBMC was detected by flow cytometry. Supernatant's Cytokines (IL - 2 and IL- 10) were assayed by ELISA. Results: The rate of apoptosis both RGH group and control group in fresh PBMC was very low and similar ( P 〉 0.05). The rate of PBMC apoptosis with RGH group was noticeably increased compared to that of normal control group ( P 〈 0.001 ) after PBMC were cultured for 48h. The level of IL- 2 was obviously lower in RGH group than in control group ( P 〈 0.(301 ), the level of IL- 10 was significantly higher in RGH group compared to control group in culture supematant (P 〈 0. 001 ). Conclusion: This study indicated that abnormal apaptosis of PBMC seriously affected Th1/Th2 type cells proliferation and may be an important mechanism of RGH patient low cellular mediated immune function, which may play an impartant role in the recurrent of GH.
