摘要
目的:探讨骨组织形成过程中骨钙素表达规律,评价金刚石-钛涂膜,纯钛和羟基磷灰石三种材料植入界面骨细胞功能情况,以利筛选出新的种植材料。方法:本实验将金刚石-钛涂膜、纯钛和羟基磷灰石三种种植材料,植入鼠下颌骨内,进行骨界面骨钙素mRNA检测及骨钙素免疫组织化学染色。结果:金刚石-钛涂膜材料新骨生成启动早,能明显加快组织的骨性愈合;金刚石和羟基磷灰石种植材料骨钙素mRNA表达量明显大于纯钛(P<0.01)。结论:骨钙素mRNA表达与界面区细胞增殖、新骨形成有关;骨钙素mRNA的检测有助于界面区细胞功能状态的判断。
Objective:To get the expression rule of osteocalcin during the formation of bone tissue. To evaluate the cell function of interface of three different materials, to get the new implant materials. Methed: three different implant materials : chemical-vapor-deposited diamond(CVD) , pure titanium (Ti) and hydroxyapatite (HA) were employed into the mandibular bone of Wistar rats. Osteocalcin mRNA was tested and immunohistochemical staining of osteocalcin was done. Result: the formation of new bone is more active and earlier in CVD diamond film than in HA and Ti materials. CVD diamond film can expedite osteointegration; the expression of Osteocalcin mRNA in the CVD and HA is much more significant than in Ti (P 〈 0.01 ). Conclusion: expression of Osteocalcin mRNA is correlated with cell increment of interface and new bone formation; Ostcocalcin mRNA examination is helpful to judge the cell function status in the interface.
