摘要
利用低温研磨、离心和连续自然沉降的方法从新鲜水稻叶片中分离得到了纯净的天然硅体,避免了传统方法中高温和强酸对硅体中有机物的破坏。由于硅体中有机物及其痕量,无法用常规生理生化方法进行化学成份分析,因此采用能谱和显微红外技术加以观测。扫描电镜和X-射线能谱分析发现硅体中除含有硅、氧元素外,还含有碳元素。进一步对硅体逐级分离后,用显微红外光谱分析,结果表明硅体中除显示多糖和少量蛋白质谱峰外,还有明显的多酚化合物的特征峰。
To understand the organic compounds associated with silica, a new method was developed to isolate pure silica bodies in fresh leaves of rice (Oryza sativa L. Var. 297) at low temperature by grinding and centtifugation procedures, which avoided the destruction of the silica body due to high temperature or strong oxidizing acids applied in the traditional method. Because there is only trace amount of organic compounds in silica bodies, they cannot be examined with conventional biochemical or bio-physiological apparatus in the laboratory. Therefore, energy dispersive X-ray spectrum and micro-infrared spectroscopy were introduced. The results showed that there was a little amount of carbon in silica bodies in addition to silicon and oxygen. Organic compounds intimately associated with silica bodies were released by treatment with HF solution. Analysis of the organic matter by micro-infrared spectroscopy revealed the possible presence of polyphenol, polysaccharide and protein.
