The treatment of melatonin on experimental knee osteoarthritis in rats

摘要 目的通过观察褪黑激素(M LT)对大鼠膝骨关节炎(OA)软骨的影响,探讨OA的发病机制及M LT不同给药方式对OA的治疗作用。方法40只W istar大鼠随机分为四组:正常对照组(A组),模型对照组(B组),M LT膝关节腔注射组(C组),M LT腹腔注射组(D组)。4%木瓜蛋白酶在1、4、7d于双侧膝关节腔各注射0.15m l建立OA模型,第8天开始D组腹腔注射M LT 1次/d,C组关节腔每3d注射M LT 1次,给药7周后处死,切取胫骨平台关节软骨大体观察,HE及甲苯胺蓝染色,M ank in评分,检测血清、关节液及滑膜中超氧化物歧化酶(SOD)、丙二醛(M DA)、一氧化氮(NO)。结果模型组关节软骨磨损,退变,软骨性骨赘形成。光镜下关节软骨表面磨损不规则,裂隙深及钙化层,部分软骨缺失,软骨细胞坏死,潮线紊乱,M ank in评分高。与B组比较,C组关节软骨形态学上明显改善,血清及关节液中SOD明显升高(P<0.01),NO、M DA均明显降低(P<0.01),而D组则未见明显改善。结论M LT能有效防治实验性OA;关节腔注射M LT治疗OA的疗效明显优于腹腔注射。 Objective: To evaluate the effects of intraartlcular and intraperitoneal injection of melatonin (MLT) on experimental osteoarthritis in rats and to research the OA pathogenesis related to free radical. Methods: Animal models of osteoarthritis in knee of rats had been established by intraarticular injection of papain at the 1th, 4th and 7th day respectivehy. The 40 rats were divided into 4 groups:unoperated animals (group A),opeatated animals(group B), OA rats that received intraartlcularl injection of MLT (group C) and OA rats that received intraperitoneal injection of MLT(group D). The animals began receving medication 1 day after the last intraarticular injection of papain and were killed 7 weeks later. Macroscopic and histologic analyses were performed on the articular cartilage. In the same time, the contents of MDA in serum and synovium, NO in serum and synovial fluid as well as the activity of SOD in serum and synovial fluid were detected. Results: Intraarticular injection of MLT alleviated the wear and degeneration of cartilage including the formation of the osteophyte, improved the morphologic appearance, decreased the Markin's marks; remarkably increased the activity of SOD in serum and synovial fluid ,lessened the contents of MDA in serum and synovium,decreased NO in synovium and synovial fluid in OA rats. Conclusions: Topical injection of MLT could delay the process of osteoarthritis by scavenging free radical. The therapeutic effects of intraarticular injection of MLT was better than that of intraperitoneal on experimental knee osteoarthritis in rats.
