Spectrophotometric Determination of Sulfamethoxazole Using Sodium 1,2-naphthoquinone-4-sulfonate as the Chemical Derivative Chromogenic Reagent

摘要 采用1,2-萘醌-4-磺酸钠体系分光光度法测定磺胺甲嗯唑(SMZ)。磺胺甲嗯唑与1,2-萘醌-4-磺酸钠在pH=10.0的缓冲溶液中发生亲核取代反应生成橙红色的产物,组成比为1:1,最大吸收波长λ=476nm;表观摩尔吸光系数ε=6.44×10^3 L/(mol·cm);SMZ浓度在0.2~60mg/L范围内呈良好的线性关系;线性回归方程为A=0.02452+0.02301C(mg/L),线性相关系数r=0.9991;检出限为0.08mg/L;RSD为0.25%(20mg/L,n=11);平均回收率为98.7%以上。优化了对磺胺甲嗯唑的测定条件。初步探讨了反应机理,应用拟定的方法测定磺胺甲嗯唑的含量,与药典法相比,结果满意。 A simple and sensitive spectrophtometric method is described for the determination of sulfamethoxazole. The method is based on a nueleophilic substitution reaction to measure the salmon pink compound produced by the reaction of sulfamethoxazole with sodium 1,2-naphthoquinone- 4-sulfonate in pH 10.0 buffer solution. The stoichiometric ratio of the compound is 1 : 1, and its maximum absorption wavelength is at 476 nm,ε=6.44 × 10^3 L/(mol· cm). The Beer's law is obeyed in the range of 0.2 -60 mg/L of sulfamethoxazole. The linear regression equation is A =0.02452 + 0. 02301C (mg/L), with a linear correlation coefficient of 0.9991. The detection limit is 0.08 mg/L, RSD is 0.3% (20 mg/L, n = 11 ), and average recovery is over 98.7%. The reaction mechanism is studied intensively. This proposed method has been successfully applied to the determination of sulfamethoxazole with satisfactory results.
