摘要
目的:探讨β转化生长因子(TGF-β)对骨缺损的修复作用。方法:TGF-β溶解后,与经化学处理后的牛松质骨载体充分混匀,真宅抽吸,冻于。将 TGF-β/载体复合物(含 TGF-β120μg、载体60mg)植入兔桡骨中上段15mm 节段性骨缺损区。设立复合物、自体骨、单纯载体及空白对照组,分别于2、4、8、12周处死动物,观察骨愈合进程。结果:术后第8周复合物及自体骨组缺损区呈骨性桥接,12周以后缺损区完全骨性修复,其密度接近正常骨,可见骨髓腔结构;单纯载体组12周时仅骨折端及尺侧骨膜有轻度反应性增生,呈骨不连征象,缺损区明显可见。结论:外源性 TGF-β对骨折及骨缺损修复有促进作用,TGF-β复合物对骨修复有潜在性临床应用前景。
Aim:To study the effects of transforming growth factorβ(TGF-β)on repairing large segmental bone de- fect.Methods:Segmental defects in the length of 1.5 cm were prepared in the mid-upper part of the radial shaft of adult rabbits.The defects were filled with implant containing TGF-βthat consisted of a carrier of 60 mg of chemically treated bovine cancellous bone and reconstituted with 120μg purified bovine TGF-β.The limbs served as the controls receiving autograrft,or carrier alone,or nothing.The defects were examined radiographically and histologically at 2,4,6 and 12 weeks after implantation.Results:In the group of implants of TGF-βcomposite,the defect areas were bridged at 8 weeks on medial and lateral sides with material Of uniform radiodensity and cortices of the cut ends were obscured by new bone.At 12 weeks after operation,the defects were bridged by uniform new bone and the cut ends of cortex could not be seen:while in the control group of single carrier,the defects remained clear.Only a small amount of new bone formed like a cap covering the cut bone ends.The appearance of the TGF-βimplanted sites in animals killed at 12 weeks showed histologically new lamellar and woven bone in the cut ends of the cortex.An early medu]lar canal appeared and contained normal-appearancing marrow elements.Conclusion:These data demonstrate that extragenous TGF-βinitiates osteogenesis and stimulates bone defects repairing in animal models,suggesting its potential clinical application in fu- ture.
