摘要 扩增含MDV主要免疫基因(gB基因)的puc19质粒并制成DNA疫苗,分别以200μg和500μg总剂量肌注免疫雏鸡,1日龄和8日龄各免疫接种一次,第二次免疫后一周,进行强毒攻击。试验期间,对死亡鸡记录并剖检。试验期结束时(99日龄),所有存活鸡全部迫杀剖检。结果显示,空白质粒对照组鸡的MD阳性率分别为84.62%(11/13),两组试验组鸡分别为15.38%(2/13)和23.08%,(3/13)。试验初步证明,MD DNA疫苗有明显的免疫保护作用。 An experiment was conducted for evaluating immunoprotectivity of Marck's disease virus (MDV)DNA vaccine. MDV gB gene was inserted into puc19 plasmid. Afterwards,chickens in two groups were intramuscularly injected with this recombi-nant plasmid DNA at age of 1 day (primary vaccination)and 8 days(boost)rcspcctively. Doses were 100 and 250μg plasmid DNA per bird each injection for two groups respectively. One week later from boost,the birds were challenged with virulent MDV strain by intra-abdominal injection. As a result, chickens with the disease, in two vaccinated groups were figured as 15. 38%and 23. 08% respectively,while the chickens bearing MD were 84. 62% in control group. The primary result shows that the DNA vaccine can effectively protect chickens against virulent MDV,and this novel vaccine strategy is very promising in development of new generation of MDV vaccine
