转基因烟草生产生物可降解塑料(PHA)遗传稳定性的初步分析 被引量:1

摘要 为检测转P.pseudoalkaligenese YS1的PHA合成酶phaC1、phaC2双价基因在转基因烟草后代植株的遗传稳定性,以已获得的两株转PHA合成酶phaC1、phaC2双价基因的烟草及其后代为材料,通过对转基因烟草各后代进行PCR、PCR-Southern检测,初步确定了P.pseudoalkaligenese YS1 PHA合成酶phaC1、phaC2双价基因能在转基因植株的后代中稳定地遗传;利用卡那霉素抗性大田直接筛选、Southern blot等方法对阳性植株作了进一步检测。用氯仿-次氯酸钠对74株PCR-Southern检测为阳性的转化植株中进行PHAs抽提,有61株中得到目的产物。所转化的烟草在产物合成水平上转化率为67.7%。
