摘要
目的检测髓系白血病细胞系HLA-Ⅱ基因型。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对4种髓系白血病细胞系(U937、K562、HEL、MEG)的HLA-Ⅱ基因型进行DNA分型。结果该4种白血病细胞系的HLA-Ⅱ基因型分别是,U937细胞:DRB1*0301/*0301,DQB1*0201/*0201,DPB1*0402/*0402;K562细胞:DRB1*0301/*0301,DQB1*0201/*0201,DPB1*0402/*0402;HEL细胞:DRB1*1302/*1302,DQB1*0201/*0201,DPB1*1401/*1401;MEG细胞:DRB1*1406/*0410,DQB1*0401/*0401,DPB1*0501/*0501。结论该4种细胞系虽然来源于不同个体,但均属于髓系肿瘤细胞,国际FAB分类均属于急性髓系白血病细胞。然而,分子免疫遗传学类型有所差异。通过本研究提高了髓系白血病细胞系HLA-Ⅱ基因型认识,对今后髓系白血病分子免疫遗传学的研究,提供了初步资料。
[Objective] To investigate the expression of HLA- Ⅱ genotype in myelogenous leukemia cell lines in order to study the relationship between HLA- Ⅱ genotype and acute myelogenous leukemia. [Methods] HLA- Ⅱ genotype of myelogenous leukemia cells lines was investigated by polymerase chain reaction - restriction fragment length polymorphism (PCR-RFLP)method. [Results] The HLA-Ⅱ genotype of K562 cells lines was as follow: DRB1 * 0301/* 0301, DQB1 * 0201/* 0201 ,DPB1 * 0402/* 0402. The HLA- Ⅱ genotype of U937 cells lines was as follow: DRB1 * 0301/* 0301, DQB1 * 0201/* 0201 ,DPB1 * 0402/* 0402. The HLA- Ⅱ genotype of HEL cells lines was as follow: DRB1 * 1302/* 1302, DQB1 * 0201/* 0201 ,DPB1 * 1401/* 1401 and The HLA- Ⅱ genotype of MEG cells lines was as follow: DRB1 * 1406/* 0410,DQB1 * 0401/* 0401 ,DPB1 * 0501/* 0501, respectively. [Conclusions] We studied the expression of HLA-Ⅱ genotype in myelogenous leukemia cell lines . The purpose of our study was to gain an increase understanding of HLA- Ⅱ genotype in myelogenous leukemia cell lines in order to study relationship between HLA-Ⅱ genotype and acute myelogenous leukemia. Further,this study is important for understanding immunogenetics of acute myelogenous leukemia in the clinic.
