摘要
应用离子交换层析法,从胎脑组织匀浆上清中提纯神经生长因子(nervesrowthfactor,NGF),其分子量约14.3KD,其3种组分的等电点分别为9.13、9.42和9.65。纯化后的NGF比活力显著提高,用鸡胚背根神经书培养法证明其具有刺激神经生长的作用。采取肌肉、球后和局部注射的方法,对45例病人进行了治疗观察。其中脑梗塞10例,脑外伤10例,外周神经损伤8例,青光眼14例,椎间盘脱出3例。总有效率为100%。NGF对大多数神经损伤表现均有作用,尤对弥漫性脑损伤、青光眼、外周神经损伤疗效最明显,未发现不良反应。
A nerve growth factor (NGF)with a MW of 14. 3KD was purified from the supernate of fetal brain tissue homogenate by ion-exchange chromatography. The isoelectric points of the 3 components of the purified NGF were 9. 13, 9. 42 and 9. 65 respectively, and the specific activity of it was significantly improved. Chick embryo dorsal root gang-lion cul tivation proved that the NGF showed the effect of stimulating nerve growth.45 cases of patients, including 10 cases of cerebral infarction, 10 cases of brain trauma,8 cases of peripheral nerve injury, 14 cases of glaucoma and 3 cases of protrusion of interver tebral disc were treated with the NGF by intramuscular, postbulbar and local injections. The total effective rate reached 100%. NGF was significantly effective in a most cases, especially those with diffuse brain injury, glaucoma and peripheral nerve injury. No adverse reaction was found.
