摘要 目的体外观察预激态补骨脂素对白血病原代细胞的杀伤作用.方法6例血标本采自住院临床确诊之白血病病例.采用体外细胞培养方法,在白血病原代细胞培养体系内加入预激态补骨脂素,使其浓度保持在0.05 g/L.短期用药实验,这一浓度维持24 h,7 d时终止实验;长期用药实验,这一浓度维持5 d,并分别在7、10、14 d时终止实验.实验均设对照组,以细胞总数、台盼蓝拒染率和台盼蓝拒染细胞抑制率等作为检测指标,以配对t检验进行组间比较.结果短期用药和长期用药均显示预激态补骨脂素对白血病原代细胞具有显著杀伤作用.培养7 d的情况下,用药时间延长对预激态补骨脂素杀伤白血病细胞作用的影响不明确.培养时间从7 d延长至14 d,台盼蓝拒染细胞抑制率从32.40%~69.50%升至94.10%~98.90%,即预激态补骨脂素杀伤白血病细胞作用得到充分表达.难治或耐药与否对预激态补骨脂素杀伤白血病细胞作用无明显影响.培养14 d时,实验组白血病细胞形态发生明显变化;而对照组基本不变,且发现少许贴壁细胞生长,实验组无此现象.结论预激态补骨脂素对白血病原代细胞具有显著的杀伤作用,有可能成为一广谱抗白血病药物. Objective To investigate the cytotoxic effects of PAP on leukemic cells in vitro. Methods Six blood samples were collected from six patients who had been diagnosed as acute leukemia clinically. PAP was added into the culture system after the culture began. The concentration of PAP in the culture system was 0.05 g/L. We kept this concentration of PAP in the culture system for 24 hours in short term PAP administration experiment where leukemic cells were counted in the 7th day. This concentration of PAP in the culture system was kept for 5 days in long term PAP administration experiment which was stoped in the 7th, 10th, and 14th day respectively. Control groups were set up and total cell counts, trypan-blue-exclusion rates, and trypan-blue-exclusion inhibition rates were checked. The statistical comparisons between the two groups were conducted through matched t tests. Resuits PAP had significant cytotoxic effects on leukemic cells in both PAP administration experiments. The influence of the prolongation of PAP administration period in the cytotoxicity of PAP towards leukemic cells was indefinite in the circumstance of 7-day culture. Trypanblue-exclusion inhibition rates of leukemic cells were elevated significantly from 32. 40%- 69. 50% to 94.10%-98. 90% when culture period was prolonged from 7 days to 14 days. The cytotoxicity of PAP was not affected by the refrectory extent of drug resistance of the cases. At the 14th day of culture the morphology of leukemic cells changed obviously in experiment group, but did not in control group. Some adherent cells grew in control group, but did not in experiment group. Conclusion PAP had significant cytotoxic effects on various leukemic cells and might become a broad spectrum anti-leukemic medicine.
