摘要
目的筛选受维甲酸调控的新基因片段,探讨这些基因诱导分化作用的机制。方法经细胞培养,构建人急性早幼粒细胞性白血病细胞的减数文库,应用半定量RT-PCR技术对减数文库的前向文库中53个新基因片段进行筛选。结果24个新基因片段受维甲酸调控,其中大多数呈上调趋势,少数表现为先下调后上调。结论维甲酸诱导的基因表达效应具有明显的时相特征。
Objective To screen novel gene segments modulated by retinoic acid and study differentiation inducing effect of the genes. Methods Using cell culture, construction of subtractive library for human's acute promyeloeytie leukemia cell and semiquantative RT-PCR technique, 53 novel gene segments were screened in the forward library of subtraetive library. Results Twenty four novel gene segments were modulated by retinoie acid, among which the majority were up-regulated, the minority were down-regulated at the beginning, then up-regulated. Conclusion Gene expression induced by retinoic acid was regularly changed with phases.
