摘要
脉冲电泳是一种可用于分离20Kb到10Mb大分子量DNA的新型凝胶电泳技术,已应用于双孢蘑菇,香菇、侧耳、草菇等多种食用菌的核型分析及核型多态性的研究。本文介绍了真菌电泳核型分析的基本操作程序和主要影响因素,并阐述了脉冲电泳在食用菌种群特异性鉴定、染色体作图和遗传分析等方面的应用前景。
Pulsed Field Gel Electrophoresis (PFGE) is a new type technique of gel electrophoresis which can be used to separate large DNA molecules ranging from 20Kb to 10 Mb. It has been applied to the electrophoretic karyotype analysis and karyotype polymorphism research of various fungi such as Agaricus bisporus, Lentinula edodes, Pleurotus spp. and Volvariella volvacea. This article describes the general manipulative procedure and primary effective factors of PFGE, and elaborates the applicable prospect of PFGE in fungal specific identification of species group, chromosome mapping and genetic analysis.
