地塞米松对大鼠肝脾细胞凋亡的影响 被引量:5

摘要 目的探讨地塞米松对大鼠肝脾等器官淋巴细胞的影响和对机体免疫抑制作用的机制。方法将20只Wistar大鼠随机分成两组,每组10只,实验组每天腹腔注射地塞米松3 mg/(kg.d),3 d后称大鼠体重、脾湿重、肝湿重。用流式细胞仪分析Annexin V/PI双染后脾细胞的凋亡率。结果实验组体重、脾湿重明显减轻(P<0.001),肝重改变不明显(P=0.75),脾细胞的凋亡率为(34.9±5.8)%,较对照组的凋亡率(0.71±0.09)%显著高(P<0.01)。结论地塞米松促进大量淋巴细胞凋亡是其抑制机体的免疫功能的一种重要机制。 Objective To study the apoptosis in the liver and spleen of rat induced by dexamethasone.Methods Dexamethasone 3 mg/(kg·day) was injected into abdominal cavity of Wistar rat for three days. The wet weight of spleen and liver was measured after dexamethasone administration. The body weight of rat before and after using dexamethasone was also measured. Apoptotic percentage of spleen cells in dexamethasone- treated rat was measured by flow cytometry using Annexin V/PI double staining method. Results The body weight of rat and the wet weight of spleen in the experimental group was significantly lower than that in control group ( P 〈 0.01).There was no significant difference in the weight of liver between two groups. Apoptotic percentage of spleen cells in experimental group was significantly higher than that in control group( P 〈 0.01 ). Conclusion Induction of lymphocyte apoptosis by dexamethasone is an important mechanism of immnunosuppression of this medcine.
