固定化黑曲霉增殖细胞生产低聚果糖的研究 被引量:10

摘要 黑曲霉孢子经海藻酸钙包埋33h后得到的固定化增殖细胞其酶活性达到最高,机械强度仍保持较高水平(为起始时80%),最适pH为5.0,与游离酶相同,最适温度为55℃,比游离酶高5℃;有害离子对其影响较小,其Km(以蔗糖为底物)为82mmol.将固定化增殖细胞装入填充式反应柱,产品中低聚果糖含量在50%~55%,操作一个月活性保持不变。 The spores of A. niger were immobilized for production of fructo-oligosaccharides.After incubation for 33 hours,the biocatalyst reached maxmum activity and mechanical stability decreased to 80% of the initial strength.The optimum pH and temperature of the immobilized growth cell were 5.0 and 55℃ respectively.The optimum PH was the same but the optimum temperature was higher by 5℃ compared with that of non-immobohjed enzyme.The immobilized beads were less sensitive to metal ions especially to Hg2+ than the non-immobohjed enzyme.The immobilized cells were packed in a column for continuous production of FOS and 50%-55% FOS(FOS/total sugar) was produced at the flow rate of lml/min. After continuous operation for more than one month,no loss of activity was observed.
