摘要 目的:建立人血浆中他扎司特的活性代谢物 MTCC 浓度的反相离子对高效液相色谱测定方法。方法:用外标法。血浆样品经酸化,用乙酸乙酯萃取后进样。色谱柱:YWG-C_(18)(4.6mm×250mm,10μm);流动相:乙腈-0.05 mol·L^(-1)磷酸盐缓冲液(1:4,pH=6.5),其中含0.004mol·L^(-1)四丁基溴化铵;流速:1.0 mL·min^(-1);紫外检测波长:254 nm。结果:血药浓度在0.02~2.00μg·mL^(-1)范围内呈良好的线性关系,r=0.9991,方法平均回收率为99.2%,日内 RSD=7.4%,日间 RSD=8.4%,平均萃取后收率为80.5%。结论:本方法操作简单,快速,灵敏度高,结果准确,重现性好,可用于他扎司特的药代动力学和生物利用度的测定。 objective:A RP - HPLC method was developed for the determintion of tazanolast's active metabolite MTCC in human plasma. Methods. The procedure involved acidulation of plasma and a single solvent extraction of drug from plasma into ethyl acetate. The column:YWG -C18 (4.6 mm 250 mm, 10 μm) ;The mobile phase:acetonitrile - 0. 05 mol · L^-1 phosphate buffer solution ( 1 : 4, pH = 6. 5 ), and 0. 004 mol · L^-1 tetrabutylammonium bromide in it. The flow rate:1. 0 mL · min^-1. The detective wave length :254 nm. Results:The regression line was linear over the plasma concentration range of 0.02 -2. 00 μg · L^-1,r =0. 9991 ,the average methodological recovery of MTCC was 99. 2%, and the average extraction recovery was 80. 5% ,the RSD of within -day and between -day was 7.4% and 8.4% ,respectively. Conclusion: :The method is simple, rapid, results is accurate and good reproducible. The analytic condition could be used to study the pharmacokineties and relative bioavailability of tazanolast.
