摘要
目的探讨胸膜成纤维细胞(FB)分泌转化生长因子-β1(TGF-β1)的作用及尿激酶(UK)对其分泌作用的影响。方法取健康雄性SD大鼠胸膜,培养、纯化并鉴定FB。设对照组和实验组。对照组分为2个亚组:对照0组由不含胸膜FB的24个样本组成,加入无血清的RPMI 1640培养液;对照1组由24个胸膜FB样本组成,加入无血清的RPMI 1640培养液,培养24 h后,用酶联免疫吸附法测定上清液中TGF-β1的含量。实验组分为4个亚组,共24个样本,分别加入5 000(A组)1、0 000(B组)、20 000(C组)和30 000 IU/mL(D组)的UK,培养24 h后取上清液,测定TGF-β1的含量。结果胸膜FB分泌TGF-β1的量为(3035.655±394.975)ng/L;A、B、C、D组(5 000-30 000 IU/mLUK)对TGF-β1均有抑制作用,A组与B、C、D组之间比较有显著性差异(P<0.01),但B、C、D组之间比较无显著性差异(P>0.05)。对照0组未检测出TGF-β1。结论胸膜FB能分泌TGF-β1,UK能抑制其分泌。
Objective To investigate the secretion of transforming growth factor beta1 (TGF-beta1) by fibroblasts(FB) and the influence of urokinase (UK) on TGF-beta1 secrection. Methods Fibroblasts obtained from the pleura of fine male rats were cultured, purified and identified, and then were divided into control group that consisted of control group 0 and control group 1 and experimental group. In control group 0, there were 24 samples without fibroblasts in RPMI 1640 cultured fluid with no serum, but in control group 1; there were 24 samples of many fibroblasts in RPMI 1640 cultured fluid with no serum; they were cultured for 24 hours and the TGF-beta1 contents in cultured suspension media were measured by enzymelinked immunoserbent assay(ELISA). Urokinase with the concentration of 5 000,10 000, 20 000 and 30 000 IU/mL respectively were added into the 24 samples in the experimental group that were divided into four sub-groups; each sub-group was cultured for 24 hours, and then the TGF-beta1 contents in cultured suspension media were measured by enzymelinked immunoserbent assay (ELISA) again. Results The concentration with which pleural fibroblasts could secret TGF-beta1 was 3035. 655 394. 975ng/L. UK with the concentration of 5 000- 30 000 IU/mL could inhibit TGF-beta1 secreting, and there was significant difference between 5 000 IU/mL UK and 10 000, 20 000 and 30 000 IU/mL UK(P〈0.01 ) ; but there was no significant difference among 10 000, 20 000 and 30 000 IU/mL UK (P〉0.05). TGF-beta1 had not been detected in control group 0. Conclusion Pleural fibroblasts can secrete TGF-beta1 and urokinase can inhibit the TGF-beta1 secretion.
