摘要
目的建立寡核苷酸阵列芯片方法,检测丙型肝炎病毒(HCV)感染后丙型肝炎患者的基因型。方法采用合成后点样的方法把自行设计合成的一系列寡核苷酸探针固定在经过修饰的显微镜载玻片上,制成用于HCV检测的基因芯片。结果对62例血清HCVRNA阳性HC患者进行基因分型,武汉地区HCV的流行株基因型主要为1b亚型(44.64%),2a亚型(19.65%)及混合型(7.14%),其余基因亚型较少见。结论采用寡核苷酸阵列芯片技术可对HCV进行基因型以及基因亚型的分析,方法快速、简便、敏感、特异。
Objective We used the oligonucleotide-microarray chip to detect the genotype of hepatitis C virus (HCV) in patients infected HCV. Methods A group of oligonucleotide probes were first synthesized, then fixed on a slide to be a microarray chip. This chip can be used for HCV detection. Results 62 serum HCV RNA-positive samples from the patients infected HCV were analyzed by the chip. The result suggested that 44. 64% of the patients were infected with HCV lb, 19. 65% with HCV 2a and 7.14% with both HCV 1 a and 2a. Conclusions This method is highly specific, sensitive and rapid for the diagnosis of the genotypes and sub-genotypes of HCV.
