用Psi2STK/GCV对鼠C6神经胶质瘤细胞作用的初步研究

摘要 目的研究利用基因工程包装细胞进行基因转导和药物治疗相结合来治疗脑胶质瘤的可行性。方法利用分子生物学方法构建了逆转录病毒载体STK及基因工程包装细胞psi2STK,用psi2STK细胞和丙氧鸟苷(GCV)结合对胶质瘤细胞进行了基因治疗研究。结果C6胶质瘤细胞和基因工程包装细胞与不同浓度的丙氧鸟苷(GCV)一起培养,未感染的细胞比感染的细胞对丙氧鸟苷(GCV)更为敏感。结论通过体外实验证实:逆转录病毒转导HSV-TK基因在培养C6胶质瘤细胞中,提高了其对GCV的敏感性。 Objective In vitro, the gene therapy of mice malignant cells was studied by the combination of psi2STK cells and ganciclovir( GCV). Methods Retroviral vector STK and package cell psi2STK was constructed by the molecular biological method. In this study, the kinetic analysis of psi2STK cells under different GCV concentrations, and sensitive analysis of C6BpBAG cells to GCV under psi2STK existence were performed. Results C6 glioma-derived cell lines and psi2STK were in culture with different concentration of GCV, compared with uninfected cells, infected cells were sensitive to GCV. Conclusion Because of being released by cell psi2STK, the replication-defective retrovirus vector with HSV-TK infected C6 cells: the sensitivity of GCV was increased. In same concentration of GCV, there was remarkable difference( P 〈 0.01 ). The study confirmed that it was possible that the. gene therapy of brain glioma, and it was an experimental basic of animal experiment in vivo and clinical usage.
