摘要
目的通过对脑胶质瘤中乙酰肝素酶mRNA的检测,探讨乙酰肝素酶在脑胶质瘤中的表达及与微血管密度的相关性。方法实时荧光定量逆转录聚合酶链反应(realtimeRT-PCR)检测47例各级人脑胶质瘤和8例正常脑组织中HPAmRNA的表达,并用免疫组化S-P法检测其微血管密度(MVD)情况,分析HPAmRNA的表达与其恶性程度及与MVD的相关性。结果胶质瘤HPA表达值随肿瘤病理级别增高而增高。不同级别组胶质瘤HPAmRNA相对值有显著性差异(P<0.01);在脑胶质瘤中HPAmRNA表达与MVD呈正相关(r=0.979,P<0.01)。结论脑胶质瘤HPA的表达与肿瘤的恶性程度、肿瘤血管生成密切相关。HPA的高表达反映了胶质瘤的恶性生物学行为,对判断预后有重要意义。
Objective To explore the expression of Heparanase (HPA) mRNA and the correlativity between it and microvessel density (MVD) in gliomas. Methods In 47 cases of gliomas and 8 cases of normal brain tissues, the expression of HPA mRNA was detected by realtime quantitative reverse transcription ploymerase chain reaction (realtime RT-PCR), and MVD was detected by immunohistochemieal technique (S-P method). The correlativity among HPA mRNA expression, malignant degree and MVD was analyzed. Results The positive expression of HPA mRNA and MVD in the gliomas were significantly higher than that in the normal brain tissues (P〈0.01). The positive expression of HPA mRNA and MVD in gliomas of grades Ⅰ - Ⅱ were significantly lower than that in the gliomas of grade Ⅲ (P〈0.01), which were significantly lower than that in the gliomas of grade IV (P〈0.01). The expression of HPA mRNA in the gliomas was positively correlated with MVD (r= 0.979, P〈0.01). Conclusions The expression of HPA mRNA in gliomas is positively related to MVD and the grade of the gliomas. The high positive expression of HPA mRNA can reflect a malignant biological behavior of glioma cells and may be of value to predicting the prognosis of the patient with glioma.
