摘要
目的:探讨抑制肌球蛋白轻链激酶(MLCK)活性诱导细胞凋亡。方法:应用尿素/甘油胶检测平滑肌细胞肌球蛋白轻链磷酸化水平,流式细胞仪检测平滑肌细胞凋亡细胞比例。结果:ML-7处理后细胞具有剂量依赖的肌球蛋白轻链去磷酸化与细胞凋亡,肌球蛋白轻链去磷酸化早于细胞凋亡。结论:抑制MLCK活性足以引起细胞凋亡,且肌球蛋白轻链去磷酸化早于细胞凋亡。
Objective To investigate the mechanism of apoptosis via inhibiting myosin light chain kinase (MLCK). Method Urea/ glycerol gel and fluorescence activated cell sorter (FACS)were used to analysis the phosphorylation /De- phosphorylation of myosin light chain (P/De-P MLC) and apoptosis,respectively. Result ML-7 caused dose-dependent dephosphorylation of myosin light chain and apoptosis in smooth muscle cells. Dephosphorylation of myosin light chain is preceded to apoptosis. Conclusion Inhibiting MLCK is sufficient to induce apoptosis, Dephosphorylation of myosin light chain is preceded to apoptosis.
