摘要
[目的]探讨胃癌组织中MMP-2(基质金属蛋白酶-2)和PTEN(张力蛋白和辅助蛋白同源、第10号染色体丢失的磷酸酶基因)蛋白的表达与其恶性生物学行为的关系。[方法]用免疫组化SP法检测64例胃癌组织和20例癌旁组织中MMP-2和PTEN蛋白的表达,并对随访的33例胃癌患者进行生存期分析。[结果]MMP-2阳性表达率在胃癌和癌旁组分别为69.0%和40.5%(P<0.05);在高中分化和低分化胃癌组分别为54.5%和48.4%(P>0.05),在侵及浆膜层和未侵及浆膜层组分别为61.5%和36.0%(P<0.05),淋巴结转移和未转移组分别为68.3%和21.7%(P<0.05),在生存期<5年和生存期≥5年组分别为75.0%和50.0%(P<0.05);PTEN蛋白阳性表达率在胃癌和癌旁组分别为77.5%和91.2%(P<0.05)。在高中分化和低分化胃癌组分别为51.5%和45.2%(P>0.05),在侵及浆膜层和未侵及浆膜层组分别为52.5%和41.7%(P>0.05),在淋巴结转移和未转移组的表达分别为26.8%和78.3%(P<0.01),在生存期<5年和生存期≥5年组分别为25.0%和58.8%(P<0.05)。胃癌组织的MMP-2与PTEN蛋白表达呈显著负相关。[结论]胃癌的MMP-2和PTEN蛋白的表达能反应其恶性潜能,并可作为判断预后的辅助指标。
[ Objective] To explore the relationship of the expressions of M M P - 2 and PTEN of its maliguant behaveirs in gastric cancer. [ Methods] The expressions of MMP- 2 and PTEN in 64 gastric carcinoma tissues were detected by immunohistochemistry SP method, and 33 patients were follavied were up. [ Results] The positive rates of M MP- 2 in carcinoma and its surrounding tissue groups were 69.0% and 40.5% respectively (P 〈 0.01) ; 54.5% and 48.2% in well and poor differentiated groups respectively ( P 〉 0.05) ; 61.5% and 36.0% in the groups with and without seraum infiltration respectively (P 〈 0.05); 68.3% and 21.7% in gastric tissues with lymph node and without lymph node metastasis; 75.0% and 58.8% in patient whose survival periods were less than 5 yeas and survival periods were more than 5 yeas. The positive rates of PTEN in carcinoma and its surrounding tissue groups were 77.5% and 91.2% respectively (P 〈 0.05); 51.5% and 45.2% in well and poor differentiated groups respectively (P 〉 0.05); 52.5% and 41.7% in the groups with and without seraum infiltration respectively (P 〉 0.05); 26.8% and 78.3% in gastric tissues with lymph node and without lymph node metastasis; 58.8% and 25.0% in patient whose survival periods were less than 5yeas and survival periods were more than 5 yeas. The expression of MMP- 2 correlated negatively with that of PTEN. [Conclusion] The expression of M MP- 2 and PTEN was closely associated with lymph node metastasis and prognosis; detecting the expression of M M P- 2 and PTEN may be helpful for predicting prognosis in gastric carcinoma.
