摘要
目的探讨单核细胞趋化蛋白-1(MCP-1)在急性胰腺炎(AP)相关的急性肺损害(ALI)中的作用。方法分别采用腹腔注射雨蛙肽和胆胰管逆行注射5%牛磺胆酸钠建立大鼠轻症、重症急性胰腺炎模型。84只SD大鼠随机分为雨蛙肽组、生理盐水组、牛磺胆酸钠组和手术对照组。用免疫组织化学法和半定量逆转录聚合酶链反应(RT-PCR)法检测各组不同时间点肺组织中MCP-1蛋白和MCP-1mRNA表达的变化情况。结果雨蛙肽组各时间点MCP-1蛋白和MCP-1mRNA的表达与生理盐水组无明显差异;牛磺胆酸钠组术后6h肺组织MCP-1蛋白和MCP-1mRNA的表达高于手术对照组(P<0.01),随着时间的延长表达逐渐增强,且与肺组织的病理改变呈正相关。结论MCP-1可能在AP相关的ALI中起了重要的作用。
[Objective] To investigate the role of Monocyte chemotactic protein-1 (MCP-1) in the pathogenesis of acute lung injury (ALl) during acute pancreatitis (AP). [Methods] AP model was induced by intraperitoneal injection of cerulein or retrograde infusion of 5% sodium taurocholate into the bili-pancreatic duct in SD rats. Eightyfour SD rats were randomly divided into four groups: cerulein group, saline control group, sodium taurocholate group and operational control group. Expression of pulmonary MCP-1 protein and MCP-1 mRNA were detected by im- munohistochemistry and semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR) at different time point in every group, respectively. [Results] No significant difference was found between cerulein group and saline control group at different time point. Expression of pulmonary MCP-1 proteins and MCP-1 mRNA in sodium taurocholate group 6 hours after operation were significantly higher than those of operational control group (P 〈0.01). Expression of MCP-1 was gradually up-regulated and correlated with the histological change of lung. [Conclusions] MCP-1 may play an important role in the pathogenesis of ALI during AP.
